2016 年,nature 上一篇《MECP2 转基因猴的类自闭症行为表征与种系传递》让他名声大噪。通过慢病毒转基因方法,首次建立了非人灵长类动物的精神疾病模型,为推动自闭症的病例与转化研究做出了重要贡献。

在仇子龙教授《一席》上的演讲中,我们能看到一个极为务实且富有人文精神的科学家,在尝试挖掘自闭症、灵长类动物模型和基因编辑之间,做出的不懈努力。

1998 年 上海交通大学生物学 学士;2003 年 中国科学院上海生命科学研究院生物化学与分子生物学 博士;2003-2009 年 加州大学 San Diego 分校神经生物学系 博士后;2009- 至今 中科院上海生科院神经所研究员、课题组长、博士生导师。

主要集中在与自闭症及瑞特综合征等发育性神经系统疾病关系密切的基因 Methyl-CpG binding protein 2 (MeCP2) 上,主要工作包括:1)发现 MeCP2 蛋白直接影响核内小 RNA 剪切加工复合物的活性而调控神经元内小 RNA 的产生,提出了发育性神经系统疾病致病机理的新观点(Cheng et al., Dev Cell, 2014),2) 发现 MeCP2 蛋白可被小泛肽化(SUMOylation)修饰而影响转录调控功能与突触发育(Cheng et la. J Neurochem, 2014),3)应用 TALEN 基因编辑方法得到敲除 MECP2 基因的食蟹猴,建立了食蟹猴基因编辑系统,为在非人灵长类中建立自闭症模型、深入研究自闭症的神经机理打下重要基础(Liu et al., Neurosci Bull, 2014)。4)应用慢病毒转基因方法得到慢病毒介导的转基因方法成功获得了 8 只存活的携带人类 MECP2 基因的转基因食蟹猴,系统性的分析了 MECP2 转基因食蟹猴的行为表型,发现 MECP2 转基因食蟹猴表现出多种自闭症患者的症状如重复刻板行为、社交障碍及升高焦虑水平等,系首次建立精神疾病的基因工程非人灵长类动物模型,为自闭症的病理与转化研究提供重要动物模型(Liu et al., Nature, 2016)。
