My laboratory has a longstanding interest in understanding the molecular mechanisms that regulate mammalian transcription and chromatin. We identified the first histone demethylase LSD1 in 2004, demonstrating the dynamic nature of histone methylation regulation, a modification that was long considered irreversible. We subsequently identified many of the second family of JmjC domain-containing demethylases, which mediate demethylation of additional lysine residues on histone tails, demonstrating that histone demethylases-mediated demethylation is a general mechanism that regulates histone methylation dynamics. We continue to investigate the mechanisms that underlie dynamic regulation of histone methylation, as well as proteins that recognize various methylated and unmethylated states of histones, and the impact on both physiological and pathological conditions.
